Haematopoiesis and Immunology (HEMATIM)
Research unit - UR 4666
HEMATIM is a young and dynamic team, created within the UPJV
in 2018. To study the regulatory mechanisms of normal and pathological immunohematopoiesis (renal failure, acute leukemia, chronic lymphocytic leukemia), HEMATIM uses fundamental models of in vitro stem cell differentiation.
Our approaches require the mastery of a large number of techniques,
including cell sorting, cytometry, biochemical studies, cloning, gene
expression modulation by lentiviral vectors, and transcriptomics by
Nanostring.
The unit’s strategy is the permanent development of innovative tools, such as genome editing by CrispR-Cas for example, or flow imaging and digital PCR.

Skills
The main themes of the unit are:
• normal and pathological erythropoiesis,
• calcium signalling in acute myeloblastic leukaemia,
• B lymphocyte differentiation and lymphoproliferative syndromes.
These themes use in vitro hematopoietic stem cell differentiation models, and are based on:
• cell sorting, flow cytometry and imaging, and immunofluorescence analyses,
• transcriptomic and proteomic analysis techniques.
• strategies for modulating the expression of genes
of interest by inducible lentiviral overexpression and knockdown by shRNA and microRNA, and genome
editing.
• Study of rare genetic diseases of erythropoiesis (Pr Garçon, Pr Da Costa, Dr Ouled-Haddou)
• Study of erythropoiesis during renal failure (Pr Metzinger and V. Metzinger-Le Meuth, MCU-HDR)
• Implications of microparticles in the physiopathology of renal transplant rejection (Dr N. Guillaume, MCUPH)
• Study of calcium signalling in acute myeloblastic leukaemia (Pr Garçon)
• Study of illegitimate Vk rearrangements in normal and pathological B lymphopoiesis (Pr Gubler)
• Study of lymphopoiesis during lymphoproliferative syndromes (Pr Marolleau)
• Functional studies of mutations associated with congenital heart rhythm disorders (Pr Rochette, Dr
Jedraszak)
• Study of the role of microRNAs in the anaemia of
chronic renal failure (Pr Metzinger and V. MetzingerLe Meuth, MCU-HDR)
Example(s) of projects
• Study of the vk-vk rearrangement in acute myeloblastic leukaemia and normal lymphopoiesis
(Dr Gubler)
• Study of the role of vitamin C in apoptosis of chronic lymphocytic leukemia cells (Pr Marolleau)
• Role of GPX4 in human terminal erythropoiesis (Pr Garçon)
• Role of the mechanotransducer PIEZO1 in normal and leukemic hematopoiesis (Pr Garçon)
• Role of microparticles in graft rejection (Dr Guillaume)
